Ronnie Moore says his squad have to be ready for a reaction from the Colchester players to the events of the week at the Weston Homes Community Stadium.
Colchester Manager Geraint Williams left his position on Monday, with Kit Symons stepping into the role on a caretaker basis, and Moore says the U's players would be out to impress.
He told Tranmere World: "We're at home to Colchester who've got a new manager. It's a completely different proposition to the one it was on Monday. Will their players get a little lift? We've got to make sure we're on our toes and play to the best of our abilities.
"They're in a false position when you look at the players they've got. They've got some good players. We're going to have to work hard and earn the right to play. If we do that, then I think we'll be ok.
"We've got to make sure we come out all guns blazing and if we do that, we're more than a match for most teams at this level."
The Manager hopes Chris Greenacre's double strike at Peterborough on Saturday will be the start of a run of goals.
"Chris scored two but could have had six," he continued. "He's a confidence player and got two goals, and he is one of those players who, when he scores one they all come together. He hit the post at Huddersfield, scored two at Peterborough and I'm sure there's plenty more to come from him."
Meanwhile, Moore was pleased to see midfielder Charlie Barnett net a hat-trick for the Reserves in their 3-0 win over Rochdale.
He added: "While we were playing 4-3-3, Charlie was in the team. But he had a little dip in form and sometimes that happens. He's going to be a hell of a player, there's no doubt about that. He has tremendous ability, great quality and can score. He's knocking on the door, which is great for me."
